If I enter a work order to make 100 ABC and this product contains a subassembly called DEF, will the system automatically create a work order to create 100 DEF? assuming 1 DEF subassembly per ABC FG.

No. It won't automatically create work orders to build your DEF.
If you create a work order to build 100 of your ABC and you 'Allocate' that work order then the shortage reports will show a shortage of your DEF reminding you to build some (or buy some).
Many people use sub contractors to build sub assemblies. Other's might build 500 in one go and put them on the shelf.

The idea of automatically generating WO's for sub assemblies wouldn't work for us.
If I make 10 product I'll create a work order to 'assemble' 10. But I don't want a work order to build 10 sub assemblies. I can't setup a machine to just make 10 subassys. I'll make a minimum of 50 but usually 100 and put them on the shelf. Later work orders to build finish product will just pull the finished sub assemblies off the shelf as required.

We are building single machines that consist of several sub-assemblies and sub-sub-assemblies. When I allocate a Work Order to a single machine, I want to see all parts from all sub-assemblies and sub-sub-assemblies with tight or negative inventory. Is there an easy option to "explode" those sub-assemblies and/or generate the necessary Work Orders for the sub-assemblies? Doing this manually for all these numerous sub-assemblies is tedious. Thanks!

If the sub assemblies are all type ASSY then the work order will consume all the components down through all levels.
If sub assemblies are type CSAS (Closed Sub Assemblies) then the work order to build the top level will assume that the CSAS sub assemblies are available so the components in those sub assemblies will not be consumed. You would normally use CSAS sub assemblies if those sub assemblies are built elseswhere or ready built on other work orders for stock.
It sounds like you want to buid everything, including the sub assemblies, inone single work order
