Is it possible to add in a comment specific to each line item added on to a purchase order?
For example i might be ordering two of the same part but one needs to be produced in plastic and one in metal.
Is it something we can use custom fields for?

Sorry no.
If the parts are made of different material then could they maybe have different part numbers/revisions. Then they could occupy separate lines on the order with different descriptions.
